Identity and purity are assessed with reversed-phase high-performance liquid chromatography, which separates the peptide from related impurities by hydrophobicity. Mass spectrometry confirms molecular weight and detects truncation or modification products. Peptide mapping after enzymatic digestion verifies the amino acid sequence. Quantitation is often performed by LC-MS/MS or by immunoassay, and the two approaches can give different values because they measure different things. Method validation parameters such as accuracy, precision, and limit of quantitation are reported alongside results.
Certificate of analysis documents from suppliers typically report purity by chromatographic area, water content, and counter-ion identity. Independent verification is advisable because reported values can be generated under differing conditions. Impurity profiles matter for research use, where aggregates, deamidation products, and residual solvents may influence experimental results. Container, lot, and chain-of-custody records support traceability. Analytical results are method-dependent, so comparisons between laboratories require the same procedure and reference standards.
Clinical development of this compound followed earlier short-acting GLP-1 analogues that required frequent injection. Once-weekly subcutaneous formulations entered use after 2017, and an oral formulation using a permeation enhancer later became available. The oral version pairs the peptide with sodium N-(8-[2-hydroxybenzoyl] amino) caprylate, a carrier that improves uptake across the gastric epithelium. Interest has expanded from glycaemic control into weight management and metabolic liver disease. Regulatory status and approved indications differ between countries, and the product remains subject to ongoing safety monitoring.
Semaglutide is a synthetic peptide that acts as an agonist at the glucagon-like peptide-1 receptor. It is a structural analogue of human GLP-1(7-37), modified to resist enzymatic degradation by dipeptidyl peptidase-4. The peptide backbone contains alpha-aminoisobutyric acid at position 8, a substitution that stabilises the helix and slows cleavage. A fatty diacid side chain attached through a linker at lysine 34 promotes binding to serum albumin, which extends the circulating half-life. These two modifications together allow less frequent administration than native GLP-1 requires.
Activation of the GLP-1 receptor couples to Gs signalling and raises intracellular cyclic AMP in pancreatic beta cells. The resulting insulin release depends on prevailing glucose concentrations, so the effect is greater when glucose is elevated. Receptor engagement also suppresses glucagon secretion and slows gastric emptying, which flattens post-meal glucose excursions. In the central nervous system, signalling in hypothalamic and brainstem regions is associated with reduced appetite and lower energy intake. Studies continue to examine effects on cardiac, renal and hepatic endpoints; whether those benefits are independent of weight change remains an open question.

The compound binds the GLP-1 receptor on pancreatic beta cells and other tissues, activating a G-protein signaling cascade that raises intracellular cyclic AMP. This action increases glucose-dependent insulin secretion when blood glucose is elevated, while binding also slows gastric emptying and reduces glucagon release. In the central nervous system, receptor activation in the hypothalamus and brainstem contributes to reduced appetite. The fatty acid chain binds albumin, which protects the peptide from renal filtration and enzymatic degradation. This albumin binding is central to its extended circulation time.
Native GLP-1 is degraded rapidly by dipeptidyl peptidase-4. Semaglutide resists this cleavage because alanine at position 8 is replaced by alpha-aminoisobutyric acid. A second substitution at position 34 introduces arginine, which further stabilizes the peptide. The most distinctive modification is a spacer and C18 fatty diacid attached at lysine 26, enabling strong albumin affinity. These three changes together produce a half-life measured in days rather than minutes, and the same structural logic underlies other long-acting analogs in this class.
Two principal therapeutic variants exist under separate regulatory filings, one indicated for glycemic control in type 2 diabetes and one for chronic weight management. Both use the same active molecule; differences lie in formulation strength, titration schedule, and labeling. Regulatory agencies in the United States and European Union approved injectable forms in 2017 and 2018 respectively. An oral tablet formulation received approval later, using a carrier molecule to enhance absorption across the gastric epithelium. Labeling differs by jurisdiction and by indication.
The distinction between established facts and open questions matters here. That the peptide binds the GLP-1 receptor and stimulates insulin release in a glucose-dependent manner is well documented. How individual variability in receptor density, gastric emptying rate, and gut microbiome composition shapes response remains an active research area. Long-term outcomes beyond five years of continuous use are not yet fully characterized in published trials, and several extension studies are ongoing.
== Use of military intelligence == Intelligence played a pivotal factor throughout the Napoleonic Wars and could very well have changed the tide of war. The use and misuse of military intelligence dictated the course of many major battles during the Napoleonic Wars. Some of the major battles that were dictated by the use of intelligence include: The Battle of Waterloo, Battle of Leipzig, Battle of Salamanca, and the Battle of Vitoria. A major exception to the greater use of superior military intelligence to claim victory was the Battle of Jena in 1806. At the Battle of Jena even Prussian superior military intelligence was not enough to counter the sheer military force of Napoleons' armies. The use of intelligence varied greatly across the major world powers of the war. Napoleon at this time had more supply of intelligence given to him than any French general before him. However, Napoleon was not an advocate of military intelligence at this time as he often found it unreliable and inaccurate when compared to his own preconceived notions of the enemy. Napoleon rather studied his enemy via domestic newspapers, diplomatic publications, maps, and prior documents of military engagements in the theaters of war in which he would operate. It was this stout and constant study of the enemy which made Napoleon the military mastermind of his time. Whereas, his opponents—Britain, Austria, Prussia, and Russia—were much more reliant on traditional intelligence-gathering methods and were much quicker and more willing to act on them.

=== Japan === In Japan, drugs and medical devices are given the designation as an orphan drug or device based on the Act of Securing Quality, Efficacy, Safety of Pharmaceuticals, Medical Devices, Regenerative or Cellular Therapy Products, Gene Therapy Products, and Cosmetics if they are intended for use in less than 50,000 patients in Japan for which there is a high medical need.
==== Crosstalk ==== Many known phosphorylation sites and O-GlcNAcylation sites are nearby each other or overlapping. As protein O-GlcNAcylation and phosphorylation both occur on serine and threonine residues, these post-translational modifications can regulate each other. For example, in CKIIα, S347 O-GlcNAc has been shown to antagonize T344 phosphorylation. Reciprocal inhibition, i.e., phosphorylation inhibition of O-GlcNAcylation and O-GlcNAcylation of phosphorylation, has been observed on other proteins including murine estrogen receptor β, RNA Pol II, tau, p53, CaMKIV, p65, β-catenin, and α-synuclein. Positive cooperativity has also been observed between these two post-translational modifications, i.e., phosphorylation induces O-GlcNAcylation or O-GlcNAcylation induces phosphorylation. This has been demonstrated on MeCP2 and HDAC1. In other proteins, e.g., cofilin, phosphorylation and O-GlcNAcylation appear to occur independently of each other. In some cases, therapeutic strategies are under investigation to modulate O-GlcNAcylation to have a downstream effect on phosphorylation. For instance, elevating tau O-GlcNAcylation may offer therapeutic benefit by inhibiting pathological tau hyperphosphorylation. Besides phosphorylation, O-GlcNAc has been found to influence other post-translational modifications such as lysine acetylation and monoubiquitination.
The loss of species from ecological communities, defaunation, is primarily driven by human activity. This has resulted in empty forests, ecological communities depleted of large vertebrates. In the Quaternary extinction event, the mass die-off of megafaunal variety coincided with the appearance of humans, suggesting a human influence. One hypothesis is that humans hunted large mammals, such as the woolly mammoth, into extinction. The 2019 Global Assessment Report on Biodiversity and Ecosystem Services by IPBES states that the total biomass of wild mammals has declined by 82 percent since the beginning of human civilization. Wild animals make up just 4% of mammalian biomass on earth, while humans and their domesticated animals make up 96%. Various species are predicted to become extinct in the near future, among them the rhinoceros, giraffes, and species of primates and pangolins. According to the WWF's 2020 Living Planet Report, vertebrate wildlife populations have declined by 68% since 1970 as a result of human activities, particularly overconsumption, population growth and intensive farming, which is evidence that humans have triggered a sixth mass extinction event. Hunting alone threatens hundreds of mammalian species around the world. Scientists claim that the growing demand for meat is contributing to biodiversity loss as this is a significant driver of deforestation and habitat destruction; species-rich habitats, such as significant portions of the Amazon rainforest, are being converted to agricultural land for meat production.

==== Greater ICE enforcement and deportations ==== Many Democratic and legal organizations have shared warnings about the expansion of immigration enforcement. Rep. Alexandria Ocasio-Cortez shared, "I don't think anyone is prepared for what they just did with ICE. This is not a simple budget increase. It is an explosion—making ICE bigger than the FBI, US Bureau of Prisons, DEA, and others combined. It is setting up to make what's happening now look like child's play. And people are disappearing."
==== Privacy Commissioner of Canada ==== In February 2023, the Privacy Commissioner of Canada, along with its counterparts in Alberta, British Columbia, and Quebec, launched an investigation into TikTok's data collection practices. In 2025, a Canadian investigation found that TikTok had collected sensitive personal data from children in violation of federal privacy laws.
== Contraindications == Contraindications of progesterone include hypersensitivity to progesterone or progestogens, prevention of cardiovascular disease (a Black Box warning), thrombophlebitis, thromboembolic disorder, cerebral hemorrhage, impaired liver function or disease, breast cancer, reproductive organ cancers, undiagnosed vaginal bleeding, missed menstruations, miscarriage, or a history of these conditions. Progesterone should be used with caution in people with conditions that may be adversely affected by fluid retention such as epilepsy, migraine headaches, asthma, cardiac dysfunction, and renal dysfunction. It should also be used with caution in patients with anemia, diabetes mellitus, a history of depression, previous ectopic pregnancy, and unresolved abnormal Pap smear. Use of progesterone is not recommended during pregnancy and breastfeeding. However, the medication has been deemed usually safe in breastfeeding by the American Academy of Pediatrics, but should not be used during the first four months of pregnancy. Some progesterone formulations contain benzyl alcohol, and this may cause a potentially fatal "gasping syndrome" if given to premature infants.

Crick was interested in two fundamental unsolved problems of biology: how molecules make the transition from the non-living to the living, and how the brain makes a conscious mind. He realised that his background made him more qualified for research on the first topic and the field of biophysics. In 1946 Crick read Erwin Schrödinger's book, What Is Life? and was influenced by it, and Linus Pauling, to switch from physics to biology. It was clear in theory that covalent bonds in biological molecules could provide the structural stability needed to hold genetic information in cells. It only remained as an exercise of experimental biology to discover exactly which molecule was the genetic molecule. In Crick's view, Charles Darwin's theory of evolution by natural selection, Gregor Mendel's genetics and knowledge of the molecular basis of genetics, when combined, revealed the secret of life. Crick had the very optimistic view that life would very soon be created in a test tube. However, some people (such as fellow researcher and colleague Esther Lederberg) thought that Crick was unduly optimistic. It was clear that some macromolecule such as a protein was likely to be the genetic molecule. However, it was well known that proteins are structural and functional macromolecules, some of which carry out enzymatic reactions of cells. In the 1940s, some evidence had been found pointing to another macromolecule, DNA, the other major component of chromosomes, as a candidate genetic molecule.
Valve is an American video game developer and publisher founded in 1996 by Gabe Newell and Mike Harrington. The company is based in Bellevue, Washington. Valve's first game was Half-Life, a first-person shooter released in 1998. It sold over nine million retail copies. Alongside Half-Life's launch, Valve released development tools to enable the player community to create content and mods. The company then proceeded to hire the creators of popular mods such as Counter-Strike. Valve continued their trend of developing predominantly first-person video games in the 2000s with a number of critically successful releases. In 2004, they released the highly anticipated sequel Half-Life 2 through their own digital distribution service Steam. The game sold over 10 million copies and was met with acclaim. Valve released two subsequent episodes for Half-Life 2 and later packaged those games together with the puzzle game Portal and the multiplayer shooter Team Fortress 2 in a collection known as The Orange Box. By the end of 2008, combined retail sales of the Half-Life series, Counter-Strike series and The Orange Box had surpassed 32 million units. Newell also projected that digital sales of Valve's games would eventually exceed retail sales as Steam continued to grow. In the late 2000s, Valve released two zombie-themed first-person shooters focusing on cooperative gameplay with the Left 4 Dead series. The company continued to release multiplayer games with the launches of Counter-Strike: Global Offensive and Dota 2, both of which have large esports communities fostered by Valve.
Vaginal estradiol is available in the forms of tablets, creams, inserts or suppositories, and rings. Vaginal estradiol tablets, creams, and inserts are usually administered once daily to twice weekly, whereas vaginal estradiol rings have a sustained action and are replaced once every 90 days. Vaginal estradiol can be used both as a systemic form of estradiol therapy, and at very low doses to selectively achieve a local vaginal effect without systemic effects, for instance in the treatment of menopausal symptoms such as vaginal atrophy and dryness. Vaginal estradiol is rapidly and almost completely absorbed. The absorption of vaginal estradiol is slightly greater in women with vaginal atrophy. Vaginal estradiol has high bioavailability and greatly increased potency compared to oral estradiol, with about 10- to 20-fold the comparative potency of oral estradiol. The greater potency of vaginal estradiol relative to oral estradiol is due to the lack of the first pass associated with oral estradiol and due to low local metabolism of estradiol in the vagina. Because of the high estradiol levels achieved, LH and FSH are more strongly suppressed with vaginal estradiol than with other routes. A daily dosage of 0.5 mg vaginal micronized estradiol has been found to result in estradiol and estrone levels of 250 pg/mL and 130 pg/mL, respectively. Vaginal estradiol has a much higher estradiol-to-estrone ratio in comparison to oral estradiol. The average ratio of estradiol to estrone with vaginal estradiol is 5:1, compared to 1:5 in the case of oral estradiol, a 10-fold difference.
On 14 April 1974, four Jewish girls were raped, murdered and mutilated after attempting to flee to Israel. Their bodies were discovered by border police in a cave in the Zabdani Mountains northwest of Damascus along with the remains of two Jewish boys, Natan Shaya, 18 and Kassem Abadi 20, victims of an earlier massacre. Syrian authorities deposited the bodies of all six in sacks before the homes of their parents in the Jewish quarter of Damascus. Syrian Sunnis were subject to severe and brutal discrimination from the Alawite-dominated Baathist apparatus; since the regime elites associated them with the Syrian opposition. As a result, Syria's Sunni community has suffered the vast majority of the brutalities and war crimes perpetrated by the Ba'athist regime during the Syrian Civil War.

The lyophilized powder is dissolved in a suitable solvent, often sterile water or a buffered diluent, with gentle mixing rather than vigorous shaking. Foaming and shear should be avoided because they can promote aggregation. The resulting solution is then stored cold and protected from light.
It usually represents the proportion of total chromatographic area attributable to the main peak. That figure does not account for impurities that do not absorb at the detection wavelength or that co-elute with the main peak. It is a useful but incomplete indicator of overall quality.
Yes. Immunoassays and chromatographic methods recognize different molecular features and can yield divergent results. Even within one technique, differences in columns, gradients, and reference standards shift reported values. Comparable numbers require a shared procedure and a common standard.
It belongs to the incretin mimetic class and acts as a long-acting glucagon-like peptide-1 receptor agonist. The class includes several peptides with different half-lives and routes of administration.
